0487 Salivary Flow Rate and cGMP in Young and Postmenopausal Women

OBJECTIVES:cGMP is important intracellular signal involved in the action of various hormones-including estrogens, neurotransmiters and vasoactive substances. On the other hand, estrogen changes may affect the composition and the flow of saliva.The aim of the present study was, therefore, to examine the relationship between the serum levels of estradiol and the salivary flow and salivary level of cGMP in women of different age. METHOD: Two groups of women were included in the study: young women (20-25 years)(7) in ovulation phase of cycle and postmenopausal women (55-60 years)(7)in good health and without of dry mouth experience. Unstimulated saliva was collected by dental cotton rolls from the floor of the mouth during 5 min. The amount of saliva was obtained after centrifugation. cGMP in the supernatant was determined by Enzyme Immunoassay for quantitative determination of cGMP (cGMP ELISA kit, Assay Designs). Concentrations of estradiol in serum were determined by chemiluminescent enzyme immunoassay (IMMULITE Estradiol, DPC) RESULTS: In group of young women, saliva flow rate, salivary cGMP and serum levels of estradiol were: 0.22 ± 0.01 ml/min, 87.20 ± 3.15 pmol/ml and 223.5 ± 10.0 pg/ml, respectively. In group of postmenopausal women, saliva flow rate, salivary cGMP and serum levels of estradiol were: 0.19 ± 0.02 ml/min, 93.60 ± 3.35 pmol/ml and 18.5 ± 3.3 pg/ml, respectively. CONCLUSION: The present findings indicate that different serum levels of estradiol in young and in postmenopausal women does not affect neither flow rate of saliva nor salivary cGMP level.
