| 2155 Effect of Mg/Zn/F-CaP on BMD of ovariectomized rats | ||

LEGEROS3, 1Musashino University, Tokyo, Japan, 2National Institute of Advanced Industrial Science and Technology (AIST), Tsukuba, Ibaraki, Japan, 3New York University, USA Mg/Zn/F-CaP (MZF-CaP) formulations consisting of carbonate apatite matrix incorporated magnesium (Mg), zinc (Zn) and fluoride (F) ions were shown to promote bone formation when used as a supplement to mineral-deficient diets (LeGeros et al, AADR, IADR 2006)or injected in ovariectomized (OVX) rats (Tokudome et al IADR 2006). Objectives: (1) Compare the efficacy of MZF-CaPs on OVX rats of two species (Sprague-Dawley, SD and Donryu), and (2) compare the efficacy when administered orally or by injection. Methods: six SD or Donryu rats were used for each of the 6 experimental groups: normal, sham control (GN), OVX treated with MZF-CaPs (G2-#51, G3-#68, G4-#76)or Zn-TCP(G1).Suspensions (10mg/0.2ml sodium arginate saline solution) of the MZF-CaP (#s 51, 68, 76) and Zn-TCP preparations were injected intramuscularly in the right thighs. #68 was also orally administered in another study. Injections or oral administrations were made once a week for 12 and 16 weeks. MZF-CaP compositions: (#51) 1.13wt% Mg, 2.91wt% Zn, 1.02wt% F; (#68) 2.63wt% Mg, 2.61 wt% Zn, 2.31 wt% F; (#76)1.94 wt% Mg, 2.86 wt% Zn and 3.16 wt%F; Zn-TCP, 6.17 wt% Zn. Bone mineral density (BMD) was measured using X-ray computed tomography (LCT-100A, Aloka, Tokyo) and bone strength measured using MX-501D (Maruto, Tokyo). Results: BMD and mechanical strengths of the femurs and vertebra were significantly higher from the treated OVX groups compared to control (untreated)for both SD and Donryu rats. Administration by injection of MZF-CaP (#68)was more effective than by oral administration in increasing BMD. Conclusion: MZF-CaP formulations are effective in improving BMD of OVX rats.
