| 2872 Antifungal Effect of nystatin vaginal tablet combined with tissue conditioner | ||

WEERAPRADIST, Mahidol University, Bangkok, Thailand Nystatin is an antifungal drug commonly use for the treatment of denture stomatitis, however, its preparation in the form of vaginal tablet has not been evaluated. Objectives: to evaluate the antifungal activity against Candida albicans of the nystatin vaginal tablet combined with a tissue conditioner during a 7-day period. The effect of incorporations of nystatin on the physical property of the tissue conditioner was also examined according to the ISO penetration test. Methods: The specimen disks (13x2 mm) of combined GC Softliner and nystatin were prepared using the concentrations of 3 times (o.6 grams) and 4 times (o.8 grams) of the minimum inhibitory concentration (MIC) value. Non-nystatin group was used as control. Specimens were contaminated with C. albicans (105 cell/ml, 50 µl) for 2 hours, then rinsed and cultured for evaluation (colonie count) of the remaining viable C. albicans. The antifungal effect was evaluated at day 0, 3 and 7, respectively. Results: The MIC of nystatin combined GC Softliner was 3,570 unit/gram. For 0.6 gram-nystatin group after specimen preparation at day 0, 3 and 7, the degree of reduction of CFU versus control were 99.9%, 99.7% and 99.6%, respectively; and the rate of negative culture were 91.7%, 66.1% and 47.2 %, respectively. For 0.8 gram-nystatin group after specimen preparation at day 0, 3 and 7, the degree of reduction of CFU versus control were similar, but the rate of negative culture were increased to 95%, 75% and 66.1 %, respectively. However, the incorporation of 0.8 gram of nystatin altered the physical property of tissue conditioner. Conclusions: It can be concluded that the combination of nystatin vaginal tablet and GC Softliner could effectively reduce high amount of Candida albicans during a 7-day period. These results suggest the possibility for the use of nystatin vaginal tablet for the treatment of denture stomatitis. | ||
