2717 Amorphous Calcium Phosphate Formation in Dentin Tubules

Objective: The sclerotic dentin in the caries dentin or the cervical defect has an advantage not only to prevent the bacterial invasion but also to decrease the dentin sensitivity because the fluid flow through the dentin tubules is interrupted by the tri-calcium phosphate crystals in the dentin tubules. We reported that the efficacy of the dentin adhesives improved in the sclerotic dentin cavity compared to that of the sound dentin. The purpose of the present study was to investigate the possibility of the formation of amorphous calcium phosphate (ACP) in the dentin tubules. Methods: The experimental material to form the artificial ACP was composed of two liquids, liquid A : aqueous mixture of potassium biphosphate and phosphoric acid, liquid B : calcium glycerophosphate solution. The liquid A followed by liquid B was applied on the flat dentin of the extracted human teeth. The morphology of the dentin tubules after the application of liquid A and B was observed by SEM. For the X-ray diffraction analysis, a disc approximately 2mm was prepared by mixing these two liquids. The analysis was performed by using X-ray diffraction apparatus (XRD 6100, Shimadzu) with Cu-K-alpha radiation. The XRD was carried out at the condition of 40kV and 30mA with a scanning speed of 5degrees/min and a scanning range of 5-85degrees. Results: By applying of liquid A for 10sec followed by liquid B for 20sec, the rod-shaped debris was observed in the dentin tubules. In addition, several peaks that indicated the tri-calcium phosphate were detected from X-ray diffraction patterns. However, the disc obtained was considered to be composed of amorphous ACP because those peaks were relatively broad. Conclusions: The artificial ACP was formed in the dentin tubules by mixing the two experimental liquids tested in this study.
