| 2556 Effects of Bone Morphogenetic Proteins on Oral Squamous Carcinoma Cells | ||

BMPs are not, however, recommended in the reconstruction of maxillofacial-skeletal defects caused by ablative cancer surgery, as their effects on residual carcinoma cells are unknown. Objective: The aim of this study was to determine the effects of BMP-2 and -7 on oral squamous carcinoma cell proliferation, and secretion of the proangiogenic factors IL-8 and VEGF. Methods: PCR was used to quantify transcripts of BMP-2, and –7, and BMP receptors Ia, Ib, II, and Act receptors I, IIa, and IIb in oral squamous carcinoma cell lines UMSCC-1, UMSCC-14 and UMSCC-74. Proliferation of the cell lines were evaluated at 24 to 96 hrs after incubation in varying concentrations of rhBMP-2 and -7 (0-500 ng/ml) or infection with an adenovirus expressing BMP-2 or -7 (AdBMP-2 and AdBMP-7, moi=0-1000). The supernatant of cells was evaluated via ELISA for expression of VEGF and IL-8. Results: BMP receptors Ia, Ib, and Act receptors I and IIa were expressed by all three cell lines. No significant increase in proliferation of any cell lines was observed with exposure to BMP-2 or -7 protein or when cells were transduced with AdBMP-2 or -7 gene (p<.05). There was no evidence of a significant increase in IL-8 or VEGF expression in any of the cell lines studied following exposure to BMP-2 or -7 protein or AdBMP-2 or -7 (p<.05). Conclusions: These results suggest that BMP proteins or BMP gene transduction does not increase the proliferation or the secretion of proangiogenic factors in oral squamous cell carcinoma cell lines in vitro. | ||
