Objectives: The purpose of this study was to evaluate the subjective and objective whitening effect containing 2.9% hydrogen peroxide in random application. Methods: Balancing for baseline color, 110 adult volunteers aged 21-32 yrs were randomized to a whitening strip containing 2.9% hydrogen peroxide polymer strip system A (thickness 390m for day time, thickness 300m for night time), a whitening strip containing 2.9% hydrogen peroxide polymer strip system B (thickness 390m for day & night time), and control group. Total contact time was 28 hours for these strip systems. Whitening was measured objectively using colorimeter (Minoltar CR-321, Japan) after 7 days trial, while tolerability was assessed through examination and using likert-scale self administrated questionnaires. Results: At the end of the treatment, adjusted mean ¥Äb* was -1.13±0.24 for the whitening strip A compared to control (p=0.01). While adjusted mean ¥Äb* was -.64±0.28 for the whitening strip B compared to control (p>0.05). Tooth sensitivity reported by 13% of subjects using the 2.9% hydrogen peroxide polymer strip A, 18% of subjects using the 2.9% hydrogen peroxide polymer strip B. The proportion of self satisfaction for whitening effect was greater 2.9% hydrogen peroxide polymer strip system A than 2.9% hydrogen peroxide polymer strip system B (72% vs 26%, p=0.010). Conclusions: Polymer strip of 300 m during night time showed subjective and objective whitening effect compared to control. On the contrary, polymer strip of 390 m during night time did not show improved whitening effect, which suggests 300 m during night time might induce more effective environment including optimal drug releasing. Detailed biochemical analysis for different oral conditions between day and night drug releasing mechanism according to polymer thickness is needed.
