| 2419 Effects of Naturopathic Supplements on Periodontal Wound Healing in vitro | ||

MADDEN, Oregon Health & Science University, Portland, USA Connective Tissue Nutrient Formula (CTNF) is a naturopathic dietary supplement designed to enhance the integrity of periodontal connective tissues. However, the efficacy and mechanisms of action of CTNF has not yet been determined. CTNF (a mixture of vitamins, minerals, and plant extracts) is currently being tested in a Phase II clinical trial as an adjunctive treatment for periodontitis. We used in vitro models of periodontal wound healing to assess the efficacy of CTNF. Objectives: To determine the effects of modifications of the CTNF formula on in vitro models of periodontal wound healing. Methods: Fibroblast mediated collagen contraction and proliferation was measured +/- various CTNF formulations. Normal human gingival fibroblast cells were grown with either a single component of CTNF or with the total CTNF formula minus a single component. Results: Vitamins A, C, D, oligoproanthocyanindins, copper, zinc, manganese, silicon, magnesium, and calcium inhibited collagen matrix contraction from 2-452%. Glucosamine sulfate and boron enhanced collagen matrix contraction by 2% and 19% (p<0.05) respectively. Vitamins A, C, D, glucosamine sulfate, oligoproanthocyanindins, copper, zinc, boron, silicon, and calcium inhibited fibroblast proliferation from 1-100%. Manganese and magnesium enhanced fibroblast proliferation by 3% and 8% respectively. CTNF minus vitamins A, C, D, glucosamine sulfate, oligoproanthocyanindins, copper, zinc, manganese, boron, silicon, magnesium, or calcium inhibited collagen matrix contraction from 513-938% and inhibited fibroblast proliferation from 59-99%. Conclusions: CTNF inhibited fibroblast mediated collagen contraction and proliferation.
