HONG3, 1Indiana University School of Dentistry, Indianapolis, USA, 2Gwangju Health College, South Korea, 3Chonnam National University, Gwangju, South Korea, 4Chonnam National University, Gwangju Various tea extracts have shown anticariogenic effects in a dose-dependent manner and oligomers of polyphenols from bark extract of the Elm tree have the ability to inhibit the activity of matrix metalloproteinases. Objectives: The objectives of this study were to investigate if Green tea and Galla Rhois extracts could inhibit plaque accumulation in an in vitro model utilizing Streptococcus mutans and whether or not they could inhibit the activity of MMPs, which are involved in degrading the dentin matrix and periodontal ligament. Methods: Methanol extracts of Green tea and Gallo Rhois (an herbal plant) were tested in vitro for their ability to inhibit the accumulation of plaque and the activity of collagenase-3 using the stainless steel wire model and a b-casein cleavage assay, respectively. Results: Methanol extracts of Green tea and Galla Rhois at 0.1% significantly inhibited the activity of MMP-3 by 75% and 79%, respectively. This was comparable to a known inhibitor of MMPs, doxycyline (75% inhibition at 0.1%). Methanol extracts of Green tea and Galla Rhois at 0.4% also inhibited in vitro plaque accumulations by 83% and 98%, respectively. Conclusion: The plant extracts tested inhibited plaque accumulation and collagenase-3 (MMP-13) in vitro. These properties of the Green tea and Galla Rhois extracts could be harnessed for the development of anti-caries and anti-periodontal treatments.
