2678 Microleakage of Five Root Canal Sealers in Dry and Wet Root Canals

Objectives: The aim of the present study was to evaluate the effect of humidity within the root canal on microleakage of root canal fillings with five root canal sealers of different material groups.

Methods: 100 extracted single rooted teeth were instrumented up to .02/#60. Removal of the smear layer was performed with 1 mL 40% citric acid, followed by 1 mL NaOCl 5% and 1 mL ethanol 70% for all teeth. Teeth were divided into ten groups of five different sealer groups for wet and dry root canals each.

Rewetting of the root canal surface was performed by storage of the teeth in a wet chamber at 100% humidity and 37°C. The root canals were filled with a single gutta-percha cone .02/#55. Linear dye penetration (linpen) was evaluated by two independent investigators using a stereo microscope. Statistical analysis was computed using SPSSwinR 10.0.

Results: Kruskal-Wallis test revealed significant differences for the ten groups (p<.001). Humidity within the root canal resulted in less microleakage for RoekoSeal, Apexit and Tubli-Seal, and higher values for AH Plus and Ketac-Endo. Thus, multifactorial ANOVA showed no influence of humidity on microleakage (p=.857) but influence of sealer (p<.001) and the combination sealer/humidity (p<.01).

Conclusions: Ketac-Endo showed the weakest performance of all sealers followed by Tubli-Seal, AH Plus, Apexit and RoekoSeal. It depends on the sealer type whether humidity has a positive or negative effect on the apical seal.
