2695 Characteristics of DNA-Lipid Films prepared by the Hot-press Method

Previously, we reported that DNA-lipid complexes and DNA-lipid films by casting of organic solution containing DNA-lipid complexes were useful candidates as dental materials. Objective: The purpose of this study was to evaluate the antibacterial activity, tensile strength and tissue response of DNA-lipid films prepared by new hot-press method. Methods: The synthesized amphiphilic lipids (C10-Gly/pts, C12-Gly/pts, C14-Gly/pts, C16-Gly/pts, C18-Gly/pts) were prepared from the reaction between Glycine, n-alkyl alcohol and p-toluenesulfonic acid. As n-alkyl alcohols, 1-decanol (C10), 1-dodecanol (C12), 1-tetradecanol (C14), 1-hexadecanol (C16) and 1-octadecanol (C18) were used. The six DNA-lipid complexes, abbreviated as DNA/C10-Gly, DNA/C12-Gly, DNA/C14-Gly, DNA/C16-Gly and DNA/C18-Gly, were easily prepared by the simple mixing from DNA and synthesized lipids. DNA-lipid complex powder was heated under press for 1 minute. As bacteria strains, Streptcoccus mutans (S.mutans) and Porphyromonas gingivalis (P.gingivalis) were used. The antibacterial activities of DNA-lipid films were evaluated by measuring inhibition zone around the disk shaped DNA-lipid films on the agar plates which were covered with bacteria. Results: The transparency DNA-lipid films were obtained by hot-press method. DNA-lipid films of DNA/C10-Gly, DNA/C12-Gly and DNA/C14-Gly showed the antibacterial activity against S.mutans. All DNA-lipid films showed the antibacterial activity against P.gingivalis. Conclusion: These results revealed that the effect of antibacterial activity of DNA-lipid films prepared by hot-press method was influenced by the length of alkyl chains in lipids.
