School of Dent. at Matsudo, Japan, 4Tokyo Institute of Technology, Yokohama, Japan Previously, we reported that the newly developed DNA-lipid complexes showed the antibacterial activity against four kinds of oral or hospital infection bacteria. Objective: The purpose of this study was to evaluate the Antifungal activity of DNA-lipid complexes against Candida albicans (C.albicans), Candida tropicalis (C.tropicalis) and Candida glabrata (C.glabrata). Methods: The amphiphilic lipids (C10-L-Ala/pts, C12-L-Ala/pts, C10-Gly/pts, C12-Gly/pts) were prepared from the reaction between amino acids (L-Alanine or Glycine), n-alkyl alcohol and p-toluenesulfonic acid. As n-alkyl alcohols, 1-decanol (C10) and 1-dodecanol (C12) were used. The four DNA-lipid complexes, abbreviated as DNA/C10-L-Ala, DNA/C12-L-Ala, DNA/C10-Gly and DNA/C12-Gly, were easily prepared by the simple mixing from DNA and synthesized lipids. The antifungal susceptibility test was performed according to National Committee for Clinical Laboratory Standards (NCCLS). The antifungal activity of DNA-lipid complexes was evaluated by the minimal inhibitory concentration (MIC:mg/ml). Results: The MICs of DNA-lipid complexes are shown in the table below.
Conclusion: These results revealed that DNA-lipid complexes showed the antifungal activity against Candida species, and the effect was influenced by the length of alkyl chains in lipids. | ||||||||||||||||||||
