| 4101 Antagonism of fluoride toxicity by high levels of calcium | ||

WANG1, 1Xinjiang Medical University, Urumqi, China, 2306 Hospital, Beijing, China Objectives: Whether the interference by fluoride (F) with human dental papilla mesenchymal cell (DPMC) could be modulated by altering the levels of calcium (Ca) and the relationship between cell activity and ALP activity in the medium was investigated. Methods: human DPMCs were exposed to 10~2630µM of F in vitro for 6~72h in the presence of high levels£¨4.1mM,10mM£Šof Ca. The activity , microstructure of the cells and secreted ALP activity were examined by MTT colorimetric analysis , TEM and enzyme kinetic method respectively. Results:The results showed that : in the presence of 263µM of F the activity of human DPMCs was inhibited and the microstructure was changed while the activity of secreted ALP was not inhibited;1315µM and 2630 µM for 12h inhibited the activity of human DPM cells wherease the activity of secreted ALP was increased;2360µM for 72h,in spite of the fact that nearly all cells were died,the activity of ALP was still high than the control. 4.1mM and 10mM levels of Ca seemed to modify the F effect, whereas the modification was not obvious when F content was 2630µM 10mM level of Ca was not more obvious than 4.1mM in modification. Couclusions:This suggested that the level of ALP was not accordant with the activity of human DPM cells;the effects of F could be partially antagonized by high Ca. | ||
